Midline signature matching in hemispheric gliomas

“Expanding the spectrum of H3K27-altered gliomas: Hemispheric cases with midline epigenetic signatures” names a setting where anatomical location and epigenetic signature labels differ. Does the paper describe how strongly the hemispheric cases match midline references relative to competing glioma signatures? I can't tell from the title whether those are partial or close matches, or whether the reference comparison accounts for age and tissue differences.

Should a partial DNA methylation match count as evidence only when its continuous score exceeds age-matched and tissue-matched variation in an independent reference population?

Does culture alter the H19 methylation reference itself?

In the reported culture-dependent decoupling of H19 allele-specific expression from H19/IGF2:IG-DMR methylation, is methylation compared with uncultured, tissue-matched reference distributions at each passage? That comparison could separate stable allelic discordance from a culture-associated shift in only part of the methylation pattern. Are passage, tissue source, and between-model variation retained in a continuous measure of that shift?

Are estimates reported separately by clock, chronological age range, blood-cell composition, and reference population? The distinction between direct and indirect genetic effects of birthweight predisposition matters for interpreting DNA methylation at birth. Beyond statistical significance, are effect sizes placed within age- and tissue-appropriate methylation reference ranges? Reporting how estimates shift across the direct, indirect and combined models could show whether adjustment removes confounding, weakens a broader signature, or changes only a small subset of sites.

For hemispheric gliomas described as having midline epigenetic signatures, what reference distribution supports the match? A convincing calibration would show score strength relative to both midline and hemispheric tumor classes, stratified by age and tissue context, rather than classify a partial resemblance as simply positive. Are anatomical discordance and uncertainty reflected in the reported match range?
