Senataxin (SETX), an RNA-DNA helicase, accumulates at transcription pause sites through the tumor suppressor BRCA1. Here, we provide mechanistic insight into how SETX-BRCA1 resolves transcription-associated R-loops to prevent deleterious outcomes.
