Spontaneous thioguanine-resistant mutants were derived from populations of finite-life-span, diploid human fibroblasts by means of a fluctuation analysis. cDNA was prepared from mutant HPRT mRNA and amplified by the polymerase chain reaction, and the sequence of the product was analyzed.
