The petite-positive yeast Saccharomyces cerevisiae can be efficiently and completely converted to respiratory-deficient cytoplasmic petite mutants by intercalating drugs. Rho0 petites from Schizosaccharomyces pombe could only be obtained in strains carrying a nuclear mutation.
