Loss-of-function mutations in NOD2, a receptor-sensing microbiota-derived muropeptides, is a genetic predisposition for Crohn's disease (CD). In this issue of Cell Host & Microbe, Gao et al. (2022) identify a microbiota-derived peptidoglycan remodeling enzyme that dampens chronic intestinal inflammation via NOD2, offering a potential path to alleviate CD.
