Total DNA was extracted using CTAB buffer from freeze-dried leaves of mature walnut (Juglans nigra) showing severe symptoms of walnut witches'-broom (WWB) disease. DNA of WWB mycoplasmalike organism (MLO) was separated from host DNA by CsCl/bisbenzimide gradient centrifugation.
