Mutants with symbiotic activity were obtained by random Tn5-Mob mutagenesis of Rhizobium meliloti 41. The place of the Tn5 insertion was localized on a cryptic middle-sized plasmid pRme41a. Mobilization of the labelled plasmid pRme41a::Tn5-Mob into R. meliloti 114, which did not contain such a plasmid led to a complete loss of the nodulation ability of the recipient strain.
