Midline signature matching in hemispheric gliomas

“Expanding the spectrum of H3K27-altered gliomas: Hemispheric cases with midline epigenetic signatures” names a setting where anatomical location and epigenetic signature labels differ. Does the paper describe how strongly the hemispheric cases match midline references relative to competing glioma signatures? I can't tell from the title whether those are partial or close matches, or whether the reference comparison accounts for age and tissue differences.

Does culture alter the H19 methylation reference itself?

In the reported culture-dependent decoupling of H19 allele-specific expression from H19/IGF2:IG-DMR methylation, is methylation compared with uncultured, tissue-matched reference distributions at each passage? That comparison could separate stable allelic discordance from a culture-associated shift in only part of the methylation pattern. Are passage, tissue source, and between-model variation retained in a continuous measure of that shift?

Age acceleration needs a clock-specific reference

For the reported association between clonal haematopoiesis and epigenetic age acceleration, the key calibration is distance from the expected methylation-age distribution, not merely a positive acceleration label. Are estimates reported separately by clock, chronological age range, blood-cell composition, and reference population? Agreement across those contexts would support a broad shift, whereas divergence could indicate a partial pattern concentrated in particular CpGs or cell fractions.

Reference ranges for divergent cortisol epigenomic patterns

How are epigenomic pattern differences between fish with divergent cortisol reactivity calibrated against age, tissue, developmental stage, and baseline population variation? A continuous measure of separation from each reference distribution would help distinguish a broad signature from a partial match driven by a subset of loci. Does the study report whether classification strength remains stable across those reference contexts?

Calibrating gestational epigenetic alterations across tissues

For epigenetic alterations associated with maternal fasting during early gestation, are effect sizes compared with gestational age and tissue specific reference distributions? The interpretation would differ if the reported pattern falls within normal developmental variation, persists across tissues, or only partially resembles a schizophrenia related signature. How are those possibilities separated, and is uncertainty in the match reported continuously rather than as presence or absence?

Separating inherited effects from methylation-signature strength

The distinction between direct and indirect genetic effects of birthweight predisposition matters for interpreting DNA methylation at birth. Beyond statistical significance, are effect sizes placed within age- and tissue-appropriate methylation reference ranges? Reporting how estimates shift across the direct, indirect and combined models could show whether adjustment removes confounding, weakens a broader signature, or changes only a small subset of sites.

Calibrating a midline signature outside the midline

For hemispheric gliomas described as having midline epigenetic signatures, what reference distribution supports the match? A convincing calibration would show score strength relative to both midline and hemispheric tumor classes, stratified by age and tissue context, rather than classify a partial resemblance as simply positive. Are anatomical discordance and uncertainty reflected in the reported match range?

Ancestry adjustment and episignature calibration

When ancestry adjustment changes an epigenome-wide signal, how should that propagate into diagnostic methylation matching? I would want match scores compared before and after adjustment within age- and tissue-matched reference distributions, with attention to whether uncertain or partial matches move across the reference range. Does the study report enough about calibration to distinguish removal of population structure from attenuation of a biologically relevant signature?
