The use of in vitro transcription of polynucleotides by prokaryotic and eukaryotic DNA-dependent RNA polymerases has proven of great usefulness in detecting mutagenic base modifications. Quite similar results are obtained with ribo- and deoxyribopolymers as templates, and with Mg2+ and Mn2+ at optimal concentrations.
