Minimal residual disease depends on what was assayed, when, and against which baseline.

Which time point defines BRAF V600 concordance?

For correlations between ddPCR liquid biopsy and radiometabolic findings, was plasma collected before therapy, during treatment, or near imaging? Please separate patients with detectable baseline BRAF V600 ctDNA from those who were never plasma-positive, and report DNA input and the assay limit for each negative sample. Otherwise, biological clearance cannot be distinguished from low shedding or a poorly timed collection.

Multicentre verification does not define every negative sample

Standardized analytical verification across dPCR and NGS platforms addresses only part of ctDNA negativity. For each site and platform, negative calls still need the tested DNA input, reportable limit at that input, variant coverage, and specimen handling window. In longitudinal testing, baseline ESR1 detectability and collection time relative to therapy determine whether absence can support molecular change rather than nonevaluability.

What anchors tumor attribution in residual disease testing?

When plasma variants are used as residual disease evidence, how are tumor-derived alterations separated from host-derived mutations? Please report whether attribution required a matched tumor baseline, paired leukocyte sequencing, or another prespecified criterion. Sampling time relative to therapy also matters: an undetected tumor-informed variant is interpretable only if it was detectable at baseline and the assay had adequate DNA input and sensitivity at the post-treatment collection.

“Clearance” requires a detectable baseline and a defined clock

In evaluating ctDNA clearance after immune checkpoint inhibition with radiotherapy, the denominator matters: patients without detectable pretreatment ctDNA cannot demonstrate molecular clearance. Post-treatment sampling should be indexed separately to immunotherapy and radiotherapy, with DNA input, assay limits, and intervening imaging reported. Otherwise, an apparent cleared state may combine biological response with low baseline shedding or an insensitive collection window.

Were BRAF ddPCR and imaging aligned in time?

For the reported clinical and radiometabolic correlatives, how closely were plasma collection and imaging paired? A low or undetected BRAF V600 signal could mean something different after therapy than before it, particularly if baseline plasma detectability was not established. Interpretation would benefit from results indexed to treatment interval, DNA input, assay sensitivity, and the collection-to-imaging gap.

A negative plasma result is conditional on assay and collection window

For standardized ctDNA ESR1 testing, cross-platform agreement alone would not establish the meaning of a negative result. Interpretation should be indexed to the mutation panel, baseline tumor genotype, plasma volume and DNA input, allele-fraction detection limits, and timing relative to therapy. In a multicentre setting, false-negative behavior should also be reported by platform and testing site. Host-derived mutations add another reason to distinguish analytical detection from tumor attribution rather than treating all plasma variants as equivalent evidence.
