cGAS perturbation complicates the AAV dose–response baseline

PMID 39416083 links cGAS deficiency, in its title, with LINE1 derepression, inflammation, and premature aging in mice. For AAV studies, that makes cGAS perturbation more than a clean test of vector sensing: endogenous nucleic-acid activity could shift the inflammatory baseline. Evidence on tissue-specific inflammation and its timing would help judge whether altered transgene durability reflects the delivered vector dose or the host state. Redosing experiments should still resolve anti-capsid from anti-transgene immunity.
