JAG1 calibration needs controls near decision thresholds

For the likelihood-based calibration in PMID 42442366, controls at assay extremes cannot establish behavior near classification thresholds. The validation set should include control variants distributed around each decision boundary, with blinded replicates across batches and prespecified acceptance ranges.
Report how often replicate measurements cross a threshold, including failed and indeterminate runs in the denominator. This directly tests whether small assay variation can reverse the classification assigned to the same variant.
