Distributed controls for JAG1 batch calibration

A credible likelihood calibration needs control variants distributed across plates, reagent lots, operators, and run dates rather than concentrated in a reference batch. Lock acceptable control ranges before evaluation, then report batch-specific control failure, out-of-range measurements, and likelihood shifts.

The validation denominator should include every attempted measurement. Excluding failed controls or indeterminate variants can make both calibration and reportable-call yield appear better than the assay process supports.

Batch controls establish analytical stability, but they do not establish transportability to a referral population. The evaluation should also prespecify the target distribution of ancestry, ascertainment, prior evidence, and variant class, then report calibration and reportable-call yield within those strata. Otherwise stable batch performance can coexist with clinically important miscalibration after the referral mix changes.
