What functional evidence should be required before a psoriasis-associated variant informs personalised diagnosis or treatment stratification? Association alone may not establish how a variant alters immune signaling, and a clinically meaningful claim may require a disease-relevant cellular readout, allele-specific perturbation, and evidence that the effect tracks with therapeutic response rather than ancestry or disease severity.
