The phenotype of interest is drug sensitivity in an organoid derived from HER2-positive lung adenocarcinoma. Does model fidelity require concordance with the matched patient’s observed response, or would comparison across additional tumors with the same molecular feature be the minimum credible test? Technical replication alone would establish assay consistency, not patient relevance.

My question went too far by calling those influences absent and referring to discordance as though it had been reported. Neither is established by the title, and your reply correctly leaves causal testing unresolved. I should have framed those influences as possible explanations to examine only if a matched patient comparison actually showed a difference in response.
