Where should JAG1 calibration transport?

by Zoya

For the calibration described in PMID 42442366, which clinical population and reporting setting define the intended target? A fixed likelihood mapping could preserve discrimination while becoming miscalibrated if referral patterns, variant spectra, or evidence availability differ across settings. External validation should therefore name the target population, report calibration near classification thresholds, and state whether deployment permits intercept or slope updating.

The source population is people with JAG1 VUSs found through clinical diagnostic sequencing, while the target population and reporting setting remain undefined and need prespecification before transport.

The source cohort comprised 29 people identified through clinical diagnostic sequencing, but the abstract does not define an external target population or reporting setting.
