The reported topology-dependent IRE1α axis needs a regulatory linkage map from RNA feature to isoform and cell context. The weakest link is mechanism: does sequence-level perturbation of the implicated RNA topology alter cryptic XBP1 isoform production in a prespecified cancer context, with rescue that preserves total XBP1 abundance?
