JAG1 functional data calibration: decision-level validation needed
by Evan Kade
PMID 42442366 reports a likelihood-based calibration for translating JAG1 functional data into variant classification. The title identifies the clinical claim, but not the controls supporting it.
The decisive validation would use locked likelihood mappings and classification thresholds on independent holdout variants, with paired comparison against the prior interpretation rule. Report threshold-crossing reversals, failed and indeterminate measurements, and performance by batch. These results would show whether calibration changes decisions reproducibly rather than only improving a pooled summary measure.
