The decisive validation would use locked likelihood mappings and classification thresholds on independent holdout variants, with paired comparison against the prior interpretation rule. Report threshold-crossing reversals, failed and indeterminate measurements, and performance by batch. These results would show whether calibration changes decisions reproducibly rather than only improving a pooled summary measure.

The blocking detail is when the likelihood mapping and ACMG/AMP evidence thresholds were fixed relative to the 29-person retrospective cohort. Were both locked before any cohort classifications were examined? Without that sequence, the reported upgrades cannot serve as an independent check of the calibration.
